today is my first lab to do my final year project- callus induction of dragon fruit in light and dark condition with different types of PGR.
i reached university at 10am...then started to prepare medium...hahahahaha although got a little bit sien....keep repeating same things....do alot of calculation...at first have to calculate how much should we take from the stock solution....
then took 2.5ml for each micro and macro elements then add PGR then added sucrose...then measured pH then added agar then topped up to 500ml then measured pH again..den done!!!!!hahahahaha
no no no..still need to autoclave it....after 1 and half hours de autoclaving still need to pour plate in laminar air flow.....
actually theres nothing much to do...just we have to wait for the laminar air flow..and the autoclave machine.....
theres only 4 laminar air flow available for PTC lab...but there are 20 of us who share the lab....
so we have to book the time haha then rebut the laminar....then we only have 3 autoclave machines....1 of that is used for decontaminate those contaminated glassware or medium (if u dare to use it to autoclave the medium just take ur own risk lo..=p) then another 2 are used for autoclave those wanted glasswares and medium lo...so we also need to rebut for it....
hahahaha everyone is rebut for it....
so...hehe i started to do at 10am then worked hard till 5pm...hahahaha geng leh..only take liao half hour de break oh..kakakakaka...
aih...sien...hope can finish prepare every medium by this week then start to sterilize and culture my dragon fruit....
u guys must pray for me....hope all my medium wont contaminate ya....kakakakaka tolong tolong.....
